CellaQuantTM Human IL-8 (Interleukin 8) ELISA Kit

Interleukin-8 (IL-8), also termed GCP-1 or NAP-1, is an 8-9 kDa CXC chemokine capable of binding heparin. It is produced by diverse cell types such as monocytes, neutrophils, fibroblasts, keratinocytes, mast cells, smooth muscle cells, dendritic cells, alveolar type II cells, and endothelial cells. IL-8 signals through two G protein-coupled receptors, CXCR1 and CXCR2, which form constitutive homodimers and can transiently heterodimerize. CXCR2, activated at low IL-8 levels, primarily mediates chemotaxis and MMP-9 release, promoting leukocyte adhesion and migration. In contrast, CXCR1 responds to high IL-8 concentrations and triggers respiratory burst and phospholipase D2 activation, enhancing neutrophil antimicrobial responses. IL-8 activity can be inhibited by complex formation with Serpin A1/alpha-1 antitrypsin, which blocks CXCR1 interaction. Beyond inflammation, IL-8 contributes to angiogenesis, atherosclerosis, and cancer progression by inducing VEGF expression, supporting endothelial cell growth, and facilitating tumor invasiveness and metastasis.

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-8. Samples (or Standards &Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-8 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-8, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Human IL-8. You can calculate the concentration of Human IL-8 in the samples by comparing the OD of the samples to the standard curve.

This ELISA kit applies to the in vitro quantitative determination of Human IL-8 concentrations in Cell culture supernatant, Cell lysates or other biological fluids.
