CRISPRa dCas9-VPR Lentiviral Vector

Lentiviral vector expressing dCas9 (D10A and H840A) fused to the VPR activator complex (VP64, p65, Rta), which enhances gene expression by recruiting transcriptional machinery. Unlike the dCas9-SAM system, dCas9-VPR does not require MS2-modified sgRNAs, simplifying delivery with a single fusion protein encoded in one vector. Using specific sgRNAs, the dCas9-VPR system can be targeted to promoter regions, enabling activation of virtually any gene of interest.
