Buffer N1

Buffer N1 is a neutralization buffer specifically formulated to rapidly restore physiological pH following alkaline lysis with Buffer B1 during plasmid or DNA purification. It typically contains potassium acetate and acetic acid, which neutralize NaOH and precipitate denatured proteins, genomic DNA, and cellular debris while allowing plasmid DNA to remain in solution. This step ensures high plasmid purity and stable supercoiled structure for downstream applications. Buffer N1 is compatible with mini, midi, maxi, and high-throughput nucleic acid purification workflows and provides consistent results across various bacterial strains.
