Taq DNA Polymerase
Taq DNA Polymerase is a thermostable enzyme derived from thermophilic organisms, enabling reliable DNA synthesis during repeated high-temperature PCR cycles. It catalyzes 5′→3′ DNA strand extension and lacks 3′→5′ exonuclease proofreading activity, making it ideal for routine amplification of DNA fragments up to approximately 5 kb. Supplied in a stabilized glycerol formulation with an optimized reaction buffer, it provides consistent performance in standard PCR workflows. One unit is defined as the amount of enzyme required to incorporate 10 nmol of dNTPs into DNA in 30 minutes at 74 °C. The enzyme also exhibits terminal transferase activity, adding a single deoxyadenosine (A) overhang to PCR products, which is useful for TA cloning. Typical applications include PCR amplification, cloning, sequencing preparation, DNA labeling, and general molecular biology assays where robustness and efficiency are required.
