Poly-Gel DNA Purification Kit

The Poly-Gel DNA Purification Kit enables efficient recovery of single-stranded or double-stranded DNA fragments directly from polyacrylamide gels (up to ~300 mg gel slice), using a silica-matrix binding system that simplifies purification and maximizes yield. After excising the DNA band, the gel slice is dissolved in a specialized buffer and passed through a filter column to remove acrylamide debris. DNA binds to the high-capacity spin column, is washed to remove salts and residual gel components, and is eluted in low-salt buffer or nuclease-free water. Typical recoveries are up to ~85% for fragments in the size range of ~50 bp to ~10 kb. The purified DNA is ready for downstream applications including ligation, PCR, Southern blotting, sequencing and cloning. Kit components include gel-dissolution/binding buffer, filter columns to remove gel debris, DNA binding spin columns, wash buffers, elution buffer and detailed protocol. No phenol/chloroform extraction or ethanol precipitation is required—ideal for high-efficiency purification from polyacrylamide gels in molecular biology, cloning and sequencing workflows.
