The process begins with mechanical disruption (e.g., grinding in liquid nitrogen) followed by chemical lysis and RNA removal, then selective binding of DNA to a maxi-spin column or magnetic-bead matrix with a binding capacity of up to ~500 μg DNA. Rigorous wash steps eliminate polysaccharides, polyphenols and other plant-derived inhibitors, and the purified DNA is eluted in low-salt buffer or nuclease-free water. The resulting DNA is ready for demanding downstream applications such as restriction analysis, long-range PCR, genotyping, sequencing and advanced genomics workflows. The kit comprises high-capacity binding columns or beads, large-volume lysis and wash buffers, RNase A, elution buffer, collection tubes and a detailed user protocol.
