The Plant DNA Midi Kit offers a streamlined workflow for purifying high-quality genomic DNA from moderate amounts of plant tissue (typically ~50–300 mg fresh/frozen tissue) or equivalent sample size. The process begins with homogenization in a lysis buffer optimized for plant metabolites (e.g., polysaccharides and polyphenols), followed by DNA binding to a mid-capacity silica-membrane column or equivalent matrix, thorough wash steps to remove inhibitors, and final elution in low-salt buffer or nuclease-free water. Typical yields range from ~10–100 μg of DNA depending on species, sample size and tissue condition. The purified DNA is highly suited for downstream applications such as PCR, qPCR, restriction digestion, Southern blotting, and sequencing. Included are the DNA binding columns (or bead format), lysis and wash buffers, collection tubes, elution buffer and a detailed user protocol.
