Calcineurin (human), (recombinant)

One U= 1pmol/min using 0.15mM RII phosphopeptide substrate (Prod. No. BML-P160) in the presence of calcium/calmodulin at 30°C. In the absence of calmodulin, the activity is <1%.

Produced in E. coli . Human calcineurin Aα (MW 60kDa) and calcineurin B (MW 15kDa) heterodimer. Both subunits are coexpressed in a construct with yeast myristoyl-CoA:protein N-myristoyltransferase. The resulting highly active calcineurin (protein phosphatase 2B) is N-myristoylated on the CaNB subunit, similar to the native protein.

Liquid. In 1X KI-128 Assay Buffer with increased DTT concentration (50mM TRIS, pH 7.5, containing 100mM sodium chloride, 6mM MgCl 2 , 5mM DTT, 0.025% NP-40 and 0.5mM CaCl 2) .

Useful for the study of enzyme kinetics, regulation, to dephosphorylate target substrates, and for screening inhibitors. Enzyme activity may be determined using the BIOMOL GREEN Calcineurin Assay Kit (Prod. No. BML-AK804) .
