GAD1 (Human) ELISA Kit
Glutamate decarboxylase 1 (GAD1) is an enzyme responsible for the synthesis of gamma-aminobutyric acid (GABA), an inhibitory neurotransmitter that plays a significant role in the central nervous system. Using pyridoxal 5′-phosphate (PLP) as a cofactor, GAD1 catalyzes the irreversible alphadecarboxylation of glutamate to form GABA and CO2. GAD1 is one of the two isoforms that exist in mammals and is primarily expressed in the brain. Together with GAD2, it maintains a constant physiological supply of GABA in the brain. Additionally, GAD1 plays an important role in the synaptogenesis and development of neurons. Reduced expression of GAD1 is associated with psychiatric disorders such as Schizophrenia, Bipolar Disorder, and Major Depressive Disorder. On the other hand overexpression of GAD1 is observed in multiple cancers such as breast, colorectal, gastric, prostate cancer, and oral SCC. GAD1 is identified as a major autoantigen in Type I diabetes, stiff-person syndrome, and epilepsy. Due to its role in multiple biological processes, GAD1 could serve as a promising target to treat human diseases. BioVision’s GAD1 (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of GAD1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of GAD1 in the sample or standard.
