Background
CD45RA recognizes Mr. 220-kdalton of the leucocyte antigen (LCA). CD45RA antigen is a member of the CD45 antigen family that also includes CD45, CD45RO, and CD45RB antigens. CD45RA is expressed on approximately 75% of CD8 lymphocytes and 50% of CD4 lymphocytes, all B cells and NK Cells. CD45RA antigen is expressed on naive T lymphocytes, antigen density decreases upon in vitro activation. A decrease antigen density has been demonstrated for CD4+CD45RA+subset with active multiple sclerosis.

Assay Principle
Add 10 µL of MAB / 100 µL of 10^6 PBMC in PBS. Mix Wash twice with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. Add 10 µL of MAB / 100 µL of Whole Blood. Mix gently and incubate for 15 minutes at room temperature 20ºC. Lyse Whole Blood and analyze or fix with 0.5% v/v paraformaldehyde in PBS and analyze. See instrument manufacturer’s instructions for Lysed Whole Blood and Immunofluorescence analysis with a flow cytometer or microscope.
