Sheep anti Mouse IgG2a (subclass specific)

The reactivity of the antiserum is directed to the subclass IgG2a. It reacts strongly withIg2a of the allelic types Igh-1a and Igh-1b, which include BALB/C, C57BL, CBA/J, SJ/J and SM/J. When used for the identification of IgG2a in other mouse strains the reaction may be weaker. The antiserum contains antibodies to iso and allospecific determinants. It does not react with other subclasses of IgG, IgG/Fab fragments, IgM and IgA or any non-Ig protein in mouse serum, as tested by immunoelectrophoresis and double radial immunodiffusion. In precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgG2a in mouse serum and other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of mouse IgG2a from serum or plasma.

Pools of purified homogenous IgG2a isolated from mouse serum of strains belonging to the allelic types Igh-1a and Igh-1b. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Precipitation assays. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells.

Delipidated, heat inactivated, lyophilized whole antiserum; No preservative added, as it may interfere with the antibody activity; Total protein and IgG concentration in the antiserum are comparable to those of pooled; Normal sheep serum; No foreign proteins added; Reconstitute the lyophilized antiserum by adding 1 mL sterile distilled water
