Background
The human LIF recombinant protein consists of a non-glycosylated polypeptide chain containing 180 amino acids. Leukemia inhibitory factor (LIF) is an interleukin 6 class cytokine involved in various developmental functions. Upon binding to its receptor (LIFR), which then associates with gp130, LIF activates JAK/STAT and MAPK signaling pathways. This activation, particularly of STAT3, is crucial for maintaining the phenotype of mouse embryonic stem cells. Produced by the endometrium, LIF plays a significant role in implantation by regulating the proliferation, invasion, and differentiation of trophoblasts. While human LIF can maintain mouse embryonic stem cells, mouse LIF is ineffective in humans due to receptor binding incompatibility. Additionally, LIF is produced by CD4+ and regulatory T cells, promoting Foxp3 expression and repressing Th17 genes, and is secreted by mesenchymal stromal cells to support hematopoiesis and immune modulation.
