CPT1A (Human) ELISA Kit
Carnitine palmitoyltransferase I (CPT1A) is an integral membrane protein that plays a significant role in the beta-oxidation of fatty acids. This liver-specific isoform of CPT is present on the outer membrane of the mitochondria of all cells except skeletal muscle cells and brown adipose cells. Unlike short-chain and medium-chain fatty acyl CoAs, long-chain fatty acyl CoAs cannot pass through the inner mitochondrial membrane. CPT1A converts these long-chain acyl CoAs to acylcarnitines, thereby enabling them to shuttle through the inner mitochondrial membrane. These acylcarnitines are re-converted to long-chain acyl CoAs in the mitochondrial matrix and undergo further metabolization to produce energy. CPT1A is less sensitive to malonyl CoA-mediated inhibition compared to other isoforms (CPT1B and CPT1C). Mutations of CPT1A lead to a rare metabolic disorder called Carnitine Palmitoyltransferase I deficiency. CPT1A is also associated with tumor development in multiple cancers such as prostate, breast, and lymphocytic leukemia. Therefore CPT1A could be a promising target to develop therapeutic agents to treat metabolic diseases and cancer. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of CPT1A in the sample or standard.
