This mRNA construct, produced viain vitrotranscription (IVT), expresses a version of theStreptococcus pyogenesSF370 CRISPR Associated Protein 9 (Cas9) protein. Cas9 is a key part of the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) genome editing system. In this system, an RNA guide sequence targets a specific site, and the Cas9 protein performs double-stranded DNA cleavage. The incorporation of two NLS signals within the mRNA increases the frequency of delivery to the nucleus, thus increasing the rate of DNA cleavage.
