CD3D & CD3E Protein (C-Flag & His, Human)

1.Flow cytometric analysis of anti-CD3 (OKT3 derived) CAR expression. 5e5 of anti-CD3 CAR-293 cells were stained with 100 μL of 5 μg/mL of AF 488-conjugated human CD3D & CD3E Heterodimer Protein (Site-Specific AF 488 Conjugation, Flag & His Tag, Cat. No. CT038-H2586H-SD) and negative control protein. Non-transduced 293 cells were used as a control (left) . AF 488 signal was used to evaluate the binding activity (QC tested) . 2.Binding activity of AF 488-conjugated CD3D & CD3E protein to PBMC cells. PBMC cells were stained with PE conjugated anti-CD3 antibody and AF 488-conjugated human CD3D & CD3E Heterodimer Protein (Site-Specific AF 488 Conjugation, Flag & His Tag, Cat. No. CT038-H2586H-SD) and detected by flow cytometry. PE signal was used to evaluate the content of CD3+ T cells in PBMCs. AF 488 signal was used to evaluate the non-specific binding activity to PBMCs (QC tested) . 3.1E5 of mouse Anti-CD3 antibody-conjugated beads (6-8 μm) were stained with a series of concentrations of AF 488-conjugated human CD3D & CD3E Heterodimer Protein (Site-Specific AF 488-Conjugation, Flag & His tag, Cat. No. CT038-H2586H-SD) . Binding activity was measured by AF 488 fluorescence. The recommended working concentration is 8 μg/mL (end-user titration is recommended for optimal performance) .
