CD28 Protein (C-His, Human)

1.Binding activity of AF 647-conjugated CD28 protein to PBMC cells. PBMC cells were stained with FITC conjugated anti-CD3 antibody and AF 647-conjugated human CD28 Protein (Site-Specific AF 647 Conjugation / His Tag, Cat. No. 11524-H86H-SG) and detected by flow cytometry. FITC signal was used to evaluate the content of CD3+ T cells in PBMCs. AF 647 signal was used to evaluate the non-specific binding activity to PBMCs (QC tested) . 2.1E5 of mouse Anti-CD28 antibody-conjugated beads (6-8μm) were stained with a series of concentrations of AF 647-conjugated human CD28 Protein (Site-Specific AF 647 Conjugation / His Tag, Cat. No. 11524-H86H-SG) . Binding activity was quantified based on AF 647 fluorescence. The recommended working concentration is 2 μg/mL (end-user titration is recommended for optimal performance) . 3.Immobilized Human, Cynomolgus, Rhesus CD28 protein — its pre-conjugation form (11524-H86H) and site-specific AF647-conjugated form (Cat. No. 11524-H86H-SG) — at 1 μg/mL (100 μL/well) can bind Anti-CD28 Antibody (Cat. No. GMP-11524-H001) with comparable EC50 values of 20-60 ng/mL (routinely tested) . 4.Immobilized Human, Cynomolgus, Rhesus CD28 protein — its pre-conjugation form (11524-H86H), site-specific AF488-conjugated form (Cat. No. 11524-H86H-SD), and site-specific AF647-conjugated form (Cat. No. 11524-H86H-SG) — at 1 μg/mL (100 μL/well) can bind Anti-CD28 Antibody (Cat. No. GMP-11524-H001) with comparable EC50 values of 20-60 ng/mL (routinely tested) .
