CD276 Protein (C-His, Human)

1.Flow cytometric analysis of anti-B7-H3/CD276 CAR expression. 5e5 of anti-B7-H3/CD276 CAR-293 cells were stained with 100 μL of 1μg/ml of AF 647-conjugated human B7-H3 Protein (ECD, AF 647 site-specific conjugation, His Tag, Cat. No. 11188-H86H-SG) and negative control protein. Non-transduced 293 cells were used as a control (left) . AF 647 signal was used to evaluate the binding activity (QC tested) . 2.Binding activity of AF 647-conjugated B7-H3/CD276 protein to PBMC cells. PBMC cells were stained with FITC conjugated anti-CD3 antibody and AF 647-conjugated human B7-H3 Protein (ECD, AF 647 site-specific conjugation, His Tag, Cat. No. 11188-H86H-SG) and detected by flow cytometry. FITC signal was used to evaluate the content of CD3+ T cells in PBMCs. AF 647 signal was used to evaluate the non-specific binding activity to PBMCs (QC tested) . 3.Flow cytometric analysis using AF 647 conjugated B7-H3 protein. 5e5 of anti-B7-H3 CAR-293 cells were stained with AF 647-conjugated human B7-H3 protein (Site-Specific Conjugation, ECD, His Tag, Cat. No. 11188-H86H-SG) . Non-transduced 293 cells were used as a control (Black Histogram) .The protein exhibited stable performance after 14 days of storage at 37°C, as measured on Day 0, Day 7, and Day 14. 4.Flow cytometric analysis using AF 647 conjugated B7-H3 protein. 5e5 of anti-B7-H3 CAR-293 cells were stained with AF 647-conjugated human B7-H3 protein (Site-Specific Conjugation, ECD, His Tag, Cat. No. 11188-H86H-SG) . Non-transduced 293 cells were used as a control (Black Histogram) . The protein exhibited stable performance after multiple freeze-thaw cycles (5, 10 cycles) .
