TNF-related Weak Inducer of Apoptosis (TWEAK) is a type II membrane protein of the tumor necrosis factor (TNF) family members, high levels expression in mouse peritoneal macrophages. The murine and human TWEAK homologs are very close related (~93% sequence identity in the receptor-binding domain) . TWEAK is a 17.3 kDa protein containing 129 residues. TWEAK induce apoptosis through cognate receptor Fn14 activate caspase-8 and caspase-3, in HSC3 cells KATO-III gastric adenocarcinoma cells and IFN-γ-treated HT-29. TWEAK stimulate inflammatory cytokines in HT-29, A375 cells, WI-38 fibroblast sand astrocytes.
