Background

Lano is a member of the LAP (leucine-rich repeats and PDZ) family of proteins that also includes Densin-180, Erbin, and hScribble. The LAP proteins generally contain multiple leucine-rich repeat (LRR) domains which serve to target them to the basolateral membrane of epithelial cells. Lano is unique in that it alone does not possess one or more PDZ (PSD95/DLG/ZO-1) domains as do the other members of the LAP family. However, it can bind to the PDZ domain of Erbin in addition to those of membrane-associated and guanylate kinase (MAGUK) proteins which regulate adhesion and plasticity at cell junctions. It has been suggested that it is through these interaction that these LAP proteins participate in the maintenance of proper embryonic development and integrity of epithelial tissues.

Function

Involved in gastrointestinal immunity. Upon stimulation by muramyl dipeptide (MDP), a fragment of bacterial peptidoglycan, binds the proximal adapter receptor-interacting RIPK2, which recruits ubiquitin ligases as XIAP, BIRC2, BIRC3, INAVA and the LUBAC complex, triggering activation of MAP kinases and activation of NF-kappa-B signaling. This in turn leads to the transcriptional activation of hundreds of genes involved in immune response. Required for MDP-induced NLRP1-dependent CASP1 activation and IL1B release in macrophages (PubMed:18511561) . Component of an autophagy-mediated antibacterial pathway together with ATG16L1 (PubMed:20637199) . Plays also a role in sensing single-stranded RNA (ssRNA) from viruses. Interacts with mitochondrial antiviral signaling/MAVS, leading to activation of interferon regulatory factor-3/IRF3 and expression of type I interferon (PubMed:19701189) .

Storage Conditions

Lano antibody can be stored at 4°C for three months and -20°C, stable for up to one year. Avoid repeated freeze-thaw cycles. Antibodies should not be exposed to prolonged high temperatures.
