Background

Collagen, type I, alpha 1, also known as COL1A1, is a human gene that encodes the major component of type I collagen, the fibrillar collagen found in most connective tissues, including cartilage. This gene is mapped to 17q21.33. And this gene encodes the pro-alpha1 chains of type I collagen whose triple helix comprises two alpha1 chains and one alpha2 chain. Type I is a fibril-forming collagen found in most connective tissues and is abundant in bone, cornea, dermis and tendon. Mutations in this gene are associated with osteogenesis imperfecta types I-IV, Ehlers-Danlos syndrome type VIIA, Ehlers-Danlos syndrome Classical type, Caffey Disease and idiopathic osteoporosis.

Activating Reagent

Aliquot 0.1 ml per well of the 4,000 pg/ml, 2,000 pg/ml, 1,000 pg/ml, 500 pg/ml, 250 pg/ml, 125 pg/ml, 62.5 pg/ml human COL1A1 standard solutions into the precoated 96-well plate. Add 0.1 ml of the sample diluent buffer into the control well (Zero well) . Add 0.1 ml of each properly diluted sample of human cell culture supernatants, serum or plasma (heparin, EDTA, citrate) to each empty well. See “Sample Dilution Guideline” above for details. It is recommended that each human COL1A1 standard solution and each sample be measured in duplicate.

Pipettes and pipette tips capable of precisely dispensing 0.5 μl through 1 ml volumes of aqueous solutions. Multichannel pipettes are recommended for large amount of samples.
