Background

Spondin 2, also known as mindin, is a protein that in humans is encoded by the SPON2 gene. SPON2 (spondin 2, mindin) is an extracellular matrix protein belonging to the mindin/F-spondin family. The protein has two amino terminal follistatin-like (FS) domains and a carboxy terminal thrombospondin type 1 repeat (TSR) domain. The FS domains mediate interactions between SPON2 and integrins, whereas the TSR domain contributes to the role of SPON2 as a pathogen pattern recognition molecule. In that capacity, SPON2 plays an important role in initiation of the innate immune response. It functions as a pattern recognition molecule within the extracellular matrix, where it binds to microbial pathogens and presents them to immune cells. Interactions between SPON2 and integrins are also critical for inflammatory cell recruitment and T cell priming. In addition to its role in innate immunity, SPON2 has also been shown to play a role in hippocampal neuronal outgrowth, cancer metastasis, and diabetic nephropathy.
