2-Guanidinobezimidazole (2GBI) is a selective agonist of NLRP3, with a KD value of 1.29 μM to His-GFP-NLRP3. 2-Guanidinobezimidazole binds directly to the LRR domain of NLRP3, promoting inflammasome assembly and activation. 2-Guanidinobezimidazole promotes anti-tumor immunity, inhibits tumor growth, and overcome immune checkpoint blockade (ICB) resistance[1].
