Talotrexin (PT523), an analog of Aminopterin, is a nonpolyglutamatable classic antifolate that specifically inhibits the reduced folate carrier (RFC), selectively impeding RFC transport and targeting dihydrofolate reductase (DHFR) to demonstrate antitumor activity and inhibit tumor growth [1] [2].
