Anti-fade Mounting Medium (With PI)

With anti-fluorescence quenching reagent, this glycerol-based mounting medium has strong effect on anti-fade and can be used to seal tissues or cells slides in fluorescence related applications. After mounting, coverslipped slides will not readily dry out and can be observed for weeks afterwards without sealing (can be stored at 4°Cor -20°Cin dark for 2-3 weeks) . The medium can prolong the duration of fluorescence and maintain the luminescence intensity of fluorescence maximally, in addition, the other components help to maintain the combination of antigen and antibody. The medium are compatible with a wide array of fluorochromes, enzymatic substrates and fluorescent proteins, such as blue fluorescent protein, FITC, Cy2, Cy3, Cy5, Cy7, DAPI, Rhodamine, Texas Red, AMCA, Hoechst 33258, Hoechst 33342, Alexa Fluor, etc. PI (Propidium Iodide, C27H34I2N4, molecular weight: 668.40 Da, CAS # 25535-16-4, purity > 95%) is an intercalating agent that can be used to stain cells. When PI is bound to nucleic acids, the fluorescence excitation maximum is 535 nm and the emission maximum is 617 nm. Excitation energy can be supplied with a xenon or mercury-arc lamp or with the 488 line of an argon-ion laser. PI is used as a DNA stain for both flow cytometry, to evaluate cell viability or DNA content in cell cycle analysis, and microscopy to visualise the nucleus and other DNA containing organelles. It can be used to differentiate necrotic, apoptotic and normal cells.
