Background
CCL21 and CCL19, the ligands for CCR7, are involved in controlling a diverse array of migratory events in adaptive immune functions such as the homing of Dendritic and T-Cells to areas of lymphoid tissues where T-cell priming occurs. These proteins play an important role in many aspects of the immune model systems, as well as general animal health. CCL19 or CCL21 induced expression in tumor tissue, from various cancer models, results in an inhibition of tumor growth and may even lead to a complete rejection of several tumors due to this increased infiltration by dendritic and/or T-cells. Both CCL19 and CCL21 are predominantly expressed in the thymic medulla and also contribute to a multitude of adaptive immune functions including thymocyte development, secondary lymphoid organogenesis, high affinity antibody responses, regulatory and memory T-cell function, and lymphocyte migration from tissues. CCL19, unlike CCL21, induces beta arrestin recruitment, which leads to receptor desensitization and internalization. Effective priming of T-cell responses depends on the interactions between naive T-cells and antigen-presenting cells (APCs) . Several animal model systems have been used to characterize the role of the CCL19, CCL21, CCR7 ligand system in T-cell responses during infection.
