Brk (PTK6) (NM_005975) Human Recombinant Protein
Product Specifications
Background
The protein encoded by this gene is a cytoplasmic nonreceptor protein kinase which may function as an intracellular signal transducer in epithelial tissues. Overexpression of this gene in mammary epithelial cells leads to sensitization of the cells to epidermal growth factor and results in a partially transformed phenotype. Expression of this gene has been detected at low levels in some breast tumors but not in normal breast tissue. The encoded protein has been shown to undergo autophosphorylation. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jan 2012]

Activity
PTK6 activity verified in a biochemical assay: PTK6 (protein tyrosine kinase 6) activity was measured in a homogeneous time-resolved fluorescent (HTRF®) assay. PTK6 is a cytoplasmic nonreceptor protein tyrosine kinase which may function as an intracellularsignal transducer in epithelial tissues. Varying concentrations of PTK6 were added to a reaction mix containing ATP and a biotinylated kinase substrate and the reaction mixture was incubated to allow the protein to phosphorylate the tyrosine residue in the substrate. HTRF detection reagents were then added, and the time-resolved fluorescent signal was measured on a Flexstation 3 microplate reader. The time resolved fluorescent signal is expressed as “delta R” or “ΔR” and is a ratio calculated from the fluorescent emission intensities of the donor and acceptor fluors.
