Background

This gene encodes a member of the ETS family of transcription factors, which are defined by the presence of a conserved ETS DNA-binding domain that recognizes the core consensus DNA sequence GGAA/T in target genes. These proteins function either as transcriptional activators or repressors of numerous genes, and are involved in stem cell development, cell senescence and death, and tumorigenesis. Alternatively spliced transcript variants encoding different isoforms have been described for this gene.[provided by RefSeq, Jul 2011]

Activity

ETS1 Activity Verified in a DNA-binding Assay: ETS1 (PH35960M5) activity was measured in a colorimetric DNA-binding assay. Purified ETS1 protein containing a C-terminal MYC/DDK tag was incubated with biotinylated double-stranded oligonucleotide containing the ETS1 consensus DNA-binding sequence. Following incubation, the reaction was transferred to a streptavidin-coated microplate to allow capture of the protein-DNA complex. After washing, the captured protein was detected with an anti-DDK peroxidase conjugate and colorimetric signal detection with TMB. Specificity of the protein-DNA interaction was confirmed by carrying out the binding in the presence of an unlabeled competitor oligonucleotide and by comparison to binding to an oligonucleotide containing a mutation in the consensus binding sequence.
