Immobilized human IL23A-His+IL12B-His at 10 μg/ml (100 μl/well) can bind human IL23R-Fc (Cat:PH51521M4) . The ECof human IL23R-Fc (Cat:PH51521M4) is 0.28-0.66 μg/ml. 3.Measured by its binding ability in a functional ELISA. Immobilized human IL23A-His+IL12B-His at 10 μg/ml (100 μl/well) can bind Cynomolgus IL23R-Fc (Cat:PK53943M4) . The ECof Cynomolgus IL23R-Fc (Cat:PK53943M4) is 0.14-0.35 μg/ml. 4. Measured by its ability to induce IL17 secretion by mouse splenocytes. The EDfor this effect is 4-20 ng/mL.

The recombinant IL23 heterodimer of human IL23A/IL12B comprises 498 (181+317) amino acids and has a calculated molecular mass of 56.3 (20.1+36.2) kDa. The apparent molecular mass of rhIL23 heterodimer is approximately 22.6 & 42.8 kDa respectively in SDS-PAGE under reducing conditions.

In general, recombinant proteins are provided as lyophilized powder which are shipped at ambient temperature. Bulk packages of recombinant proteins are provided as frozen liquid. They are shipped out with blue ice unless customers require otherwise. Stable for 1 year at -20°C or below from date of shipment. For maximum recovery of product, centrifuge the original vial after thawing and opening the cap. For long-term storage, aliquot and store at -20°C or below. Avoid repeated freeze-thaw cycles.

A DNA sequence encoding the IL23A (NP_057668.1) (Met1-Pro189) was fused with a polyhistidine tag at the C-terminus, constructed the plasmid 1; A DNA sequence encoding the p40 subunit of human IL12, termed as IL12B (NP_002178.2) (Met1-Ser328) was fused with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the IL23 heterodimer was purified.
