SAE1 protein, forming a heterodimer, acts as an E1 ligase for SUMO1, SUMO2, SUMO3, and potentially SUMO4. It critically mediates ATP-dependent activation of SUMO proteins, enabling the formation of a thioester bond with UBA2/SAE2's active site cysteine.
