Background
RNF43, or ring finger protein 43, is an E3 ubiquitin ligase that acts a tumor suppressor to negatively regulate the WNT ((wingless-related integration site) signaling pathway through promoting the degradation of frizzled WNT receptors. Loss of function mutations in RNF43 have been shown to be a predictive biomarker in favorable response to anti-BRAF ((B-Raf proto-oncogene serine/threonine kinase) /EGFR ((epidermal growth factor receptor) therapies in colorectal cancer patients. RNF43 mutations are frequent predictors, and therefore of high clinical interest, in pancreatic, gastric, and endometrial cancers. Further studies will reveal more on the cancer signaling pathways and effective combinatorial therapies involving RNF43.
