This anti-CD3 antibody is a purified recombinant antibody, which is labeled with fluorescein isothiocyanate (FITC) . This antibody has been tested by flow cytometry for specific binding to human CD3 on the surface of PBMCs (BPS Bioscience #79059) .FluoSite™is a tag-based method for site-specific conjugation of antibodies with fluorophores, ensuring the labeling site is far from the antigen-binding domain to preserve antibody functionality. This precise, consistent labeling ensures the homogeneity of the labeled antibody preparation, making it ideal for flow cytometry and other high-sensitivity applications.

Background

CD3 (cluster of differentiation 3) is a complex made of CD3γ chain, a CD3δ chain and two CD3ε chains. CD3 associates with the TCR (T-cell receptor) and a CD3ζ chain to activate cytotoxic T Cells (CD8+) and T helper cells (CD4+) and form the TCR complex. It is found in T cells, during all developmental stages, and it is a typical T cell marker. Its essential role in T cell activation has made CD3 an attractive and useful molecule for the development of anti-cancer drugs. For example, CAR-T cells (chimeric receptor antigen T cells) made use of the CD3ζ domain for intracellular signaling activation.

Cross Reactivity

This antibody recognizes human CD3. It has not been tested on other species.

Label

Fluorescein isothiocyanate (FITC) is a derivative of fluorescein and has excitation and emission spectrum peak wavelengths of approximately 495 nm and 519 nm, causing it to fluoresce green.

Applications

Useful for studying expression of CD3 on the cell surface by flow cytometry. For flow cytometric staining, the suggested use of this reagent is 5 µl (1 µg antibody) per million cells in 100 µl staining volume.
