This anti-CD123 antibody is a purified human recombinant antibody, which is labeled with R-Phycoerythrin (PE) . This antibody has been tested by flow cytometry for specific binding to human CD123 on the surface of CD123 CHO Recombinant cells (BPS Bioscience #79640-H) .FluoSite™is a tag-based method for site-specific conjugation of antibodies with fluorophores, ensuring the labeling site is far from the antigen-binding domain to preserve antibody functionality. Background

CD123, also known as interleukin 3 receptor alpha (IL3Ra), is a transmembrane protein found in pluripotent progenitor cells and involved in hematopoietic lineage differentiation. It can also be found in basophils, plasmacytoid dendritic cells and dendritic cells. CD123 is overexpressed on leukemic hematopoietic stem cells (HSCs), but not normal HSCs. It has been found in acute myeloid and B-lymphoid leukemias, blastic plasmacytoid dendritic neoplasms (BPDCN) and hairy cell leukemia. This expression profile makes CD123 an attractive target in cancer therapy. CAR-Ts and other therapies targeting CD123 are currently being evaluated in clinical trials.

Label

PE is an oligomeric protein complex (270 kDa) from red algae that exhibits intensely bright red-orange fluorescence with excitation peak at 566 nm and an emission peak at 574 nm. The complex consists of six heterodimers, α subunit (18 kDa) and β-subunit (20 kDa), and an additional γ-subunit (34 kDa) .

Applications

Useful for studying expression of CD123 on the cell surface by flow cytometry. For flow cytometry staining, the suggested to use this reagent at 5 µl (1 µg antibody) per million cells in 100 µl staining volume
