Background

CDC7 (cell division cycle 7-related protein kinase), also known as DDK (Dbf4/Drf1-dependent kinase), is a serine/threonine kinase involved in cell cycling. Once activated by binding to DbF4b it phosphorylates MCM2 (minichromosome maintenance 2), a protein of the MCM helicase complex, and DNA synthesis is initiated. It is also crucial for the activation of ATR (ataxia telangiectasia and RAD3 related protein) and CHK1 (checkpoint kinase 1) . It is involved at the S-phase checkpoint, allowing for controlled and faithful DNA replication. It is found at high levels in cancer cell lines and tumors, where it can contribute to cell cycle deregulation and genome instability. Knockdown of this protein in cancer cell lines leads to cell death, via an ATR-dependent mechanism. Studies in Ewing sarcoma, the second most common bone cancer in children and teenagers, showed that inhibition of CDC7 leads to mitotic catastrophe and increased DNA damage. The development of inhibitors targeting CDC7 is an ongoing area of research, with the goal of improving specificity and reducing off-target effects.
