PAK6 (p-12-activated kinase 46 is a member of the PAK family of serine/threonine kinases. The PAK family is highly evolutionary conserved and regulates cell survival, apoptosis, cytoskeleton dynamics, and other critical cellular functions. PAK6 belongs to group II, which can be activated by Cdc42 (cell division control protein 42) only as opposed to group I, which depend on either Rac1 or Cdc42.I It is ubiquitously expressed, participating in cytoskeleton remodeling, cell motility, apoptosis, and metastasis. It has been found at high levels in prostate and colon cancer and hepatocellular carcinoma. It has been correlated with resistance to cancer therapy with 5-flurouracil, docetaxel and radiation. Recently it has been shown that PAK6-related resistance to oxaliplatin treatment is due to an increase in HR (homologous recombination) DNA repair, by translocating from the cytoplasm to the nucleus and activating ATR (ataxia telangiectasia and Rad-3 related protein) and CHK1 (checkpoint kinase 1) . The development of specific inhibitors targeting PAK4, to be used alone or as combination therapy, will greatly benefit the cancer therapy field.
