LGP2 protein is a key regulator of RIGI- and IFIH1/MDA5-mediated antiviral signaling and has dual negative and positive functions affected by factors such as infected viruses and target cells. LGP2 lacks a CARD domain for independent initiation of antiviral signaling and therefore inhibits RIG-I through multiple mechanisms, including competition for RNA binding and obstruction of interaction with MAVS/IPS1.
