To study the roles of calcium, a key intracellular messenger in cells, it is essential to quantitatively monitor its concentration. The most widely used method of Ca2+ detection is the use of fluorescent Ca2+ indicators. Rhod-590 AM ester is a membrane-permeant calcium indicator that can be loaded into cells via incubation.. Rhod-590 AM ester itself does not bind Ca2+, but it is readily hydrolyzed to Rhod-590 by endogenous esterases once it enters cells. Experiments using indicators in cells usually are performed within one or two hours of loading, but it may be possible to re-load cells with indicator if needed.
