RMC-4791 is a RAS (ON) multi-selective tri-complex inhibitor (TCI) that binds the KRAS switch II region differently from daraxonrasib, with reduced dependence on the Y64 interaction. This enables RMC-4791 to retain inhibitory activity against KRAS Y64 resistance mutants and effectively suppress ERK signaling and cell proliferation, highlighting its potential to overcome daraxonrasib resistance.
