CV2a is a 14-3-3-directed molecular glue-based PROTAC that selectively targets the ERα protein complex, with an EC50 of 0.10 μM. It facilitates the recruitment of VHL to the 14-3-3/ERα complex and enhances the potent polyubiquitination of the 14-3-3ζ/ERα complex.
