EP4 receptor antagonist 7 is a potent and selective small molecule antagonist of the PGE2 EP4 receptor (IC50 = 1.1 nM) . It effectively suppresses PGE2-mediated signaling and gene expression in macrophages in vitro and demonstrates synergistic antitumor activity with anti-PD-1 therapy in mouse colon cancer models by enhancing CD8+ T cell infiltration.
