STAT3-IN-38 (Compound 4m) is a celastrol-derived small molecule that binds the STAT3 SH2 domain with a K_D of 45.33 μM, inhibiting Tyr705 phosphorylation. This action suppresses Survivin and Mcl-1 expression, leading to cell cycle arrest and apoptosis in colorectal cancer cells in vitro.
