SOS1-IN-17 (Compound 8d) is a potent, orally bioavailable SOS1 inhibitor that disrupts the SOS1-KRASG12C interaction (IC50 = 5.1 nM) . It effectively inhibits ERK phosphorylation and cellular proliferation in KRASG12C-mutant cancer cells and demonstrates in vivo antitumor activity in pancreatic cancer mouse models.
