TAS-121 is a potent, covalent third-generation EGFR-TKI that selectively targets mutant EGFR forms, including T790M resistance mutations, and inhibits HER2/4. It demonstrates robust antitumor efficacy in cellular assays and in vivo xenograft models, such as NCI-H1975, by blocking EGFR phosphorylation and downstream signaling.
