NACE2i
NACE2i is a cell-permeable peptide inhibitor specifically targeting nuclear ACE2 without affecting importin pathway-regulated nuclear proteins. It selectively disrupts IMP1α-ACE2 binding, thereby inhibiting SARS-CoV-2 viral replication while preserving ACE2 enzymatic activity. In preclinical models, NACE2i suppresses inflammation and macrophage infiltration while enhancing NK cell infiltration in bronchioles. Treatment elevates H3K27ac levels (an active histone mark), restores host translation in infected bronchiolar cells, and increases methylated ACE2 in bronchioles and lung macrophages. With dual mechanisms, NACE2i both curbs lethal inflammation associated with severe COVID-19 and limits viral replication by restoring host translation of antiviral cytokines.
