BBO-8520 is a first-in-class dual-state KRASG12C inhibitor that covalently binds to the Switch-II pocket, blocking signaling in both ON and OFF conformations. It demonstrates potent activity in growth factor-stimulated in vitro assays and induces durable tumor regression in vivo, including in models resistant to OFF-state-only inhibitors.
