JTT-654 is a potent, selective, and orally active inhibitor of 11β-HSD1, with low nanomolar IC50 values for human, rat, and mouse enzymes and high selectivity over 11β-HSD2. It functions as a competitive inhibitor and has been studied in research models for its potential to ameliorate insulin resistance and non-obese type 2 diabetes by targeting adipose and hepatic tissues.
