KIN-8194 is a potent, orally bioavailable dual inhibitor of HCK and BTK with sub-nanomolar IC50 values. It demonstrates activity against BTK inhibitor-resistant malignancies, including mantle cell lymphoma, by inhibiting proliferation and adhesion, and has shown in vivo efficacy in overcoming ibrutinib resistance in DLBCL xenograft models.
