KOR agonist 3 (Compound 5) is a potent and selective κ opioid receptor (κOR) agonist (EC50 = 0.88 nM) with significant selectivity over μOR (EC50 = 720 nM) and no δOR activity below 1 μM. It demonstrates biased signaling with minimal β-arrestin-2 recruitment compared to U50488, making it a valuable research tool for studying κOR-mediated neuropharmacology in vitro and in vivo.
