HDAC8-IN-12 (compound 5k) is a potent and selective non-hydroxamic acid HDAC8 inhibitor (IC50 = 0.12 nM) . It demonstrates anti-breast cancer activity in vitro and in an orthotopic mouse model, where it modulates the tumor immune microenvironment by activating T cells and shifting macrophage polarization from M2 to M1.
