INCB16562 is a selective, orally active JAK1/2 inhibitor that potently blocks IL-6-induced STAT3 phosphorylation in myeloma cells. It overcomes cytokine and stromal protection, sensitizing cells to chemotherapeutics in vitro and demonstrating anti-tumor efficacy in mouse xenograft models, supporting its research use in hematological malignancies.
