U0126 is a potent and selective noncompetitive inhibitor of MEK1 and MEK2 (IC50: 72 nM and 58 nM, respectively), blocking the MAPK/ERK pathway. It suppresses AP-1 activity and has been used in vitro to inhibit the anchorage-independent growth of ras-transformed cells and in vivo to study cancer and neurological disease models.
